The following year, the young physician served as a Medical Resident in Radiology at the Massachusetts General Hospital, a position he held until 1965. In addition, Mattoso worked as a Professor at the School of Radiological Techniques. In April 1964, he was designated as the physician responsible for the Diagnostic Radiology and Radiotherapy Department to attend the Oak Ridge Institute for Nuclear Studies medical center.
